چکیده

Eucaryotic cells contain two complete genetic systems, one based on nuclear DNA and the other on cytoplasmic DNA . The study of these systems has been greatly facilitated by the existence of specific inhibitors . One such inhibitor is the trypanocidal drug, ethidium bromide (Short, Peak, and Watkins, 1953) . Application of the drug to trypanosomes in vitro causes alterations in the structure of their kinetoplast (mitochondrial) DNA (Riou and Delain, 1969) . In HeLa cells, ethidium bromide (EtBr)l inhibits the synthesis

The Effect of Ethidium Bromide on Mitochondrial Dna Synthesis and Mitochondrial Dna Structure in Hela Cells

The synthesis of mitochondrial DNA (mDNA) in HeLa cells is selectively inhibited by relatively low concentrations of ethidium bromide. After exposure of cells to strongly inhibitory concentrations of the drug, the apparent superhelix density of mDNA is rapidly increased, as judged by its buoyant density in CsCl in the presence of ethidium bromide. Mitochondrial DNA synthesized in the presence o...

Inhibition of phytohaemagglutinin--induced lymphocyte transformation and mitosis by ethidium bromide.

The effect of ethidium bromide (EB) on phytohaemagglutinin (PHA) induced lymphocyte transformation and mitosis was evaluated by morphology and 3H-thymidine uptake. EB at low concentrations is known to specifically bind to mitochondrial DNA and selectively inhibit mitochondrial protein synthesis. However at these concentrations EB inhibited both transformation and mitosis. Electron micrographs o...

Mitochondrial Protein Synthesis in Hela Cells

HeLa cell mitochondrial proteins have been shown to be the products of two separate protein-synthesizing systems; one, the general cellular mechanism, sensitive to inhibition by cycloheximide, the other, a specific mitochondrial system subject to inhibition by low concentrations of chloramphenicol (Galper, J. B., and J. E. Darnell. 1971. J. Mol. Biol 57:363). Preliminary data have suggested tha...

Insertional editing of mitochondrial tRNAs of Physarum polycephalum and Didymium nigripes.

tRNAs encoded on the mitochondrial DNA of Physarum polycephalum and Didymium nigripes require insertional editing for their maturation. Editing consists of the specific insertion of a single cytidine or uridine relative to the mitochondrial DNA sequence encoding the tRNA. Editing sites are at 14 different locations in nine tRNAs. Cytidine insertion sites can be located in any of the four stems ...

Electroporation of DNA into Physarum polycephalum Mitochondria: Effects on Transcription and RNA Editing in Isolated Organelles

Mitochondrial RNAs in the acellular slime mold Physarum polycephalum contain nucleotides that are not encoded in the mitochondrial genes from which they are transcribed. These site-specific changes are quite extensive, comprising ~4% of the residues within mRNAs and ~2% of rRNAs and tRNAs. These "extra" nucleotides are added co-transcriptionally, but the means by which this is accomplished have...
